How to back up the thread database?

1.) How can I back up my FILESTREAM database on SQL Server 2008 from SQL Server Management Studio?
2.) Do I have to make a separate backup of the FILESTREAM folder on the disk?

1.) You can backup the stream data using the regular SQL Backup and recovery process. See this article for reference. You can also exclude filter data from an explicit backup if you wish.

2.) You can take a separate backup of the filestream directory, but you really don't need it. This only makes sense for the instance of the SQL Server database that created it.
